Description
Kick Them Out!!! (KTO) is the only semi-coherent spin-off of the Eurogamer PlayTime feature Kick the Bricks. The player controls a ball and, using wall kicks, try to kick out as many wooden bricks as possible. While the idea of kicking a ball out of your way is simple enough, it takes time to build up the huge bricks that slowly make up the levels, and a constant need to avoid obstacles.
Some levels, such as Level 9, are very basic, with the ball simply attempting to bounce off the top of the level, but most involve the player using the ball to help him or her avoid dangerous obstacles. Several times during the game, randomly determined segments of a level are removed, forcing the player to restart the level at the last checkpoint. During these times, the player must avoid the removed segments without the balls being thrown into the void to the left or right of the level.
As well as keeping the bricks safely out of reach, the player must also avoid obstacles such as the floating egg halves (which explode if they hit the bricks) and a sinkhole located at the start of each level.
It is also possible to hit bricks which have already been kicked out. If a brick is kicked out that hits a brick to the left or right of the level, then a piece of the brick is removed, requiring the player to kick it out again, leading to a third brick being removed.

The targeting of various ligands, particularly hormones and growth factors, to specific cells is essential for their biological activity. However, the mechanisms underlying receptor-mediated targeting, the internalization and recycling of endocytosed ligands, the direct delivery of ligands to intracellular domains, and the intracellular sorting of ligands following internalization remain poorly understood. The present study reports the use of isolated perfused rat liver to characterize the stability of receptor-mediated targeting of ligands (insulin) on liver cells. Under steady state conditions, we found that the initial targeting of 125I-insulin to isolated hepatocytes occurred in two steps, with half-times of 6.5 and 21 min. By comparing the initial and total targeting of 125I-insulin, we estimated that one-third of the receptors are internalized and trafficked through the Golgi complex, whereas the remainder of receptors are shed back into the medium. Internalized ligand is subsequently rapidly cleared from the endosomal compartment. Although 125I-insulin has been shown to be internalized by an energy-dependent process with a t1/2 of approximately 3 min, we found that a substantial pool of the internalized ligand appeared to remain within the endosomal compartment (t1/2 = 0.5 h). These data suggest that the internalization and trafficking of ligands through the endosomal compartments to the lysosomal compartment may be the essential pathway for the internalization and sorting of ligands in the liver.
