More information about the pack or visiting  to access full version.Heterotetrameric complex of wild type and mutant lamin-B1 associated with laminopathy and lipodystrophy: identification of two novel mutant lamin-B1s.
Laminopathies include a group of rare genetic disorders of the skeletal muscle that are characterized by muscle wasting and weakness. Characteristic histological features include a well-circumscribed vacuolar degeneration of muscle fibers associated with intranuclear and cytoplasmic invaginations of the nuclear envelope termed inclusions. A major component of the inclusions are the lamin-B proteins and most genetic mutations in the lamin-B1 gene are associated with forms of the diseases termed "progeroid laminopathies." Recent studies of lamin-B1-deficient mice indicated that neither the wild type nor the mutant lamin-B1 protein are toxic to cells. Here we demonstrate that both wild type lamin-B1 and two mutant lamin-B1 alleles resulting in laminopathies can associate in the same cell, a heterotetrameric complex in vitro. The in vivo distribution of these heterotetramers was then studied. The wild type lamin-B1 formed heterotetramers with two mutant lamin-B1s in normal skeletal muscle and in muscles from the patients. In addition, the mutant lamin-B1 proteins self-associated forming multimers and abnormal oligomers. These data indicate that normal and mutant lamin-B1 can interact, and that these interactions lead to the formation of pathologic heterotetramers.The following is a transcript of the video interview I did with Jennifer Jacobs for Forbes on March 26, 2018.
