The effect of sustained exercise training on cardiac fibrosis, left ventricular hypertrophy, and cardiomyocyte size in the early phase of pressure overload (PO) hypertrophy was examined in conscious, chronically instrumented Sprague-Dawley rats. Five weeks after a constrictor band was placed around the abdominal aorta, the animals were randomly assigned to either a sedentary or an exercise group and submitted to a 1-mo regime of treadmill running (10 m/min, 20 min/d, 5 days/wk). Exercise significantly reduced left ventricular and interstitial fibrosis, but had no effect on cardiomyocyte size. At 5 wk of PO, cardiac fibrosis was associated with upregulated expression of the myofibroblast marker alpha-smooth muscle actin (alpha-SMA) and focal deposition of matrix proteins and transforming growth factor-beta1 (TGF-beta1).
