The main character Shizu is a kid living with his mother at their rundown apartment. One night a loud noise alerts his mother and she goes to the window to see what it is. There is a grey fog that slowly swallows her until all that’s left of her is Shizu, who is still crying.
Pumpkin Eater: All About Shizu is the complete horror game spin-off that tells the story of the boy who sees his mother swallowed by the fog. The game was created by the authors of Pumpkin Eater, with scenarios written by Shizue. It is available as an application or on the web.

Shizu is a young boy who starts to feel hungry one night. He is looking for food in his mother’s cupboard when he hears a suspicious sound and suddenly the sound is followed by a thick grey fog. Mom is gone, and Shizu wonders where she went. He hears some sounds in the kitchen and follows them to the living room, where his mother is. He sees her sleeping in the corner. Shizu doesn’t know what to do and where to look for his mom, and as he falls asleep he hears a noise from the kitchen. In the morning he finds a note: “I’m taking the train to my parents’. Give my things to Dad and don’t try to find me.”
Pumpkin Eater: All About Shizu is a free-to-play browser game that can be played online. Players are in control of Shizu, who decides whether to move the furniture to open up the kitchen, try to hunt for his mother, use the washing machine and other household appliances, or even try to fall asleep. The house has limited amount of resources, which can be earned in various ways and can be used to upgrade the apartment. Players also have a store where they can buy items for themselves or give away Shizu’s food.

Glial-derived neurotrophic factor (GDNF) regulates the development of cerebellar granule neurons (CGNs) and enhances synapse formation and neural plasticity of CGNs. The molecular mechanisms of GDNF action are mostly unknown. Here we show that GDNF induces the rapid phosphorylation of its target transcriptional repressor, downstream regulatory element of amyloid precursor protein-constitutive fragment (DREAM), in CGNs. Phosphorylation of DREAM was increased by GDNF and was decreased by inhibition of AKT, a key molecule in the AKT signaling pathway. The DREAM phosphorylation was shown to be important for the regulation of gene expression by DREAM. GDNF enhances expression of c-fos and BDNF in CGNs. A CRE, a sequence in the c-fos and BDNF promoter, is necessary for GDNF-induced BDNF expression. The BDNF CRE is overlapped with CREB-binding protein/p300 binding sites. Phosphorylated CREB/p300 interacts with DREAM as shown by in vitro and co-immunoprecipitation experiments. In CGNs, phosphorylated DREAM interferes with the interaction between CREB/p300 and CRE, thereby allowing CREB-mediated gene expression. A DREAM-like protein identified in CGNs also bound to CREB/p300. Taken together, our results suggest that DREAM is a key molecule in GDNF-induced gene expression in CGNs.
