The locking solution you're looking for is called Shared Access Protection, or SAS.  Essentially, if you have files that are shared by multiple users, you create a special group of users for that purpose, and each time a user is granted permission to the file, they're added to the group, rather than being given permissions by their IP.  A single user can only open the file once, but as soon as they're granted permission, the file is open and there's no need for them to have another request.
This is actually implemented in modern Windows editions, and is a built-in part of Windows 7 and Windows Server 2012.  It also works in Linux (Fedora at least).  I wrote a small app, not available, but published here for various versions of Windows.  You can probably get the source and a compiled version for a specific edition of Windows somewhere.
In Windows Server 2008, I've heard that this is not necessarily enabled by default.  I don't know if it's still the case, but SAS isn't turned on by default in that version.  I'll search it up and edit this answer.
By default, the program works on files, which means it won't work for folders.  You can get around that by implementing some sort of blocking dialog, by checking the size, etc, and see whether or not the file is open and is greater than 1mb.  Or, you can use the fact that Microsoft itself uses such a scheme, and implement a special file that checks itself against the list in the file to see if it is open or not.  Or, you can just use a regular file with a size of one.
If you want to allow subfolders, you'll have to come up with a different solution (other than checking the filesize or something).
