‎.‎Down-regulation of heparin cofactor 2 by heparin treatment or senescent cells increases vascular smooth muscle cell growth and induces inflammatory responses.
Heparin cofactor 2 (HC2) is an essential cofactor for thrombin-mediated activation of protease-activated receptor 1 (PAR1) and mediates smooth muscle cell (SMC) growth. Recent studies demonstrate the presence of senescent cells in atherosclerotic plaques. To investigate the involvement of HC2 in senescent cells, we treated vascular SMCs with heparin, a potent inhibitor of HC2. We found that, in contrast to senescent cells, heparin treatment did not affect the expression of HC2 and PAR1. On the other hand, proliferation of SMCs was dramatically enhanced by heparin treatment. The growth promotion of SMCs by heparin was abolished by a knockdown of HC2 expression with a specific small interfering RNA. We also show that HC2 is involved in anti-inflammatory effects of heparin. In macrophages and vascular SMCs, heparin suppressed the expression of inflammatory genes, including CCL2, CXCL2, IL-8, and NF-κB, and IL-1β and IL-6 production. The effects of heparin on the expression of inflammatory genes were abolished by HC2 knockdown in SMCs. These findings reveal that heparin treatment or senescent cells induces SMC proliferation by inhibiting HC2 expression, and that HC2 is involved in heparin-mediated suppression of inflammatory gene expression in macrophages and SMCs.Q:
