An easy way to have a single instance of your program is to have it link with a library named "SingleInstance" (this program is the single instance) and then be added to the 'Startup' group of the program which will load first.
This works for Win32 and all others. Just don't forget to remove the entry from the registry (if it's installed) or rename the program's manifest file. 
The author of the library is very good at documenting how to do this and it seems he is not available on the Internet.

There is an easier way to do this that I found recently (and seem not to have been posted in a while).
Firstly, the term Single Instance which is used to describe "a program that runs as a single instance and doesn't share the memory with any other instances" seems to have been deprecated by Microsoft.
It seems that people use the term instead to describe "a program that is started from the start menu and does not appear to run in any other windows until the first user clicks on the start menu. In this case, the start menu is the "instance" of the application.
To achieve this, start the executable of your "instance" program like normal. Then, after it finishes, go to "Programs and Features" in the Windows Control Panel, and check if the program is listed under "Run this program as an administrator". If the program
