The MAPK and apoptosis pathways are involved in stress-induced changes in salivary secretion of young and aged rats.
Stress has been associated with alterations in salivary secretion. Previously we showed that the response of salivary secretion to stress is greater in aged than young rats. Since stress-induced salivary secretion is mediated by neurotransmitter receptors, the aim of this study was to evaluate the participation of neurotransmitter receptors in this phenomenon. For this, we evaluated the participation of MAPK and apoptosis pathways on salivary secretion in young (7 months) and aged (20 months) rats submitted to stress in the presence or absence of the neurotransmitter receptors antagonists. Animals were submitted to stress by electrostimulation of the superior cervical ganglion (SCG) for 1h, and salivary flow rate was determined 30 min before the stimulation and after 1h. The inhibition of neurotransmitter receptors or inhibition of MAPK and caspases (apoptosis) pathways was achieved with the administration of yohimbine (5mg/kg), prazosin (3mg/kg) and SB203580 (20mg/kg), respectively. Salivary flow rate was increased in young rats after SCG stimulation. The same pattern was observed in aged rats with the increase of salivary flow rate. In addition, the administration of yohimbine, prazosin and SB203580 to aged rats reduced salivary flow rate. In young rats, only the inhibition of the MAPK pathway produced the same effects. In aged rats, MAPK and caspases inhibition did not produce the same result, since salivary flow rate was reduced in the presence of caspases inhibition.
